Study of Enzyme Kinetics

A standard solution of substrate (300 mg/l) starch solution was taken.

Different concentrations of substrate were taken while keeping enzyme concentration constant. Volume was mad up to 2ml using distilled water. O.5 ml of enzyme (amylase) solution was added in every t.t. and incubated at 370C for 20 min. Reaction was stopped by keeping the tt in ice-bath. Samples were analyzed for glucose concentration using DNS method.

Result and Discussion: From the graph the value of Vmax & Km is determined and found to be as –

Vmax     =    44.44 µ moles / min.
                        Km          =    171 mg/l

With increase in initial substrate concentration the value of product formation increases sharply initially and after sometimes it becomes constant. Which can be explained by Michaelis – Menton equation.
