Determination of Volumeteric Mass Transfer Co-efficient (Oxygen balance method)

The animation describes the bioreactor normally operating under batch cultivation mode (As described in detail earlier). The reactor is equipped with flat blade turbine impeller and the air is purged from the disc sparger. Temperature of the reactor is controlled by circulating constant temperature water through the jacket. The broth pH is controlled by addition of acid/alkali through peristaltic pump. The dissolved oxygen of the fermentation broth is being measured by Dissolved Oxygen probe. While the gaseous oxygen concentration is measured by the paramagnetic oxygen analyzers installed in the inlet/exit air lines.

Start the experiment by pressing “Reset button” Different points are generated in a graph of CL vs (Fi – Fo) primarily depending on the % air supply and air being absorbed by the growing culture in the bioreactor. The real time values are seen in the window on the right hand side. The plotting of the graph stops when Fi = Fo A straight line connecting all the points is generated by the system From the slope of the line of the plot CL vs (Fi – Fo) KLa can be calculated as ρa (density of the air), Q (Volumetric flow rate of the air), V (Volume of the reactor) is known. The intercept of above graph gives C*.
